Functions in nuclear protein import as an adapter protein for nuclear receptor KPNB1. Binds specifically and directly to substrates containing either a simple or bipartite NLS motif. Docking of the importin/substrate complex to the nuclear pore complex (NPC) is mediated by KPNB1 through binding to nucleoporin FxFG repeats and the complex is subsequently translocated through the pore by an energy requiring, Ran-dependent mechanism. At the nucleoplasmic side of the NPC, Ran binds to importin-beta and the three components separate and importin-alpha and -beta are re-exported from the nucleus to the cytoplasm where GTP hydrolysis releases Ran from importin. The directionality of nuclear import is thought to be conferred by an asymmetric distribution of the GTP- and GDP-bound forms of Ran between the cytoplasm and nucleus.

KPNA6 also known as importin alpha 7 functions in the nuclear import of proteins. It acts by forming a complex with importin beta allowing proteins to pass through the nuclear pore complex. KPNA6 has a molecular mass of approximately 60 kDa. Expression of KPNA6 occurs in several tissues including liver brain and testes emphasizing its widespread role in various physiological processes.
KPNA6 facilitates the translocation of proteins containing a nuclear localization signal into the nucleus. It is part of the classical importin alpha/beta complex where it binds cargo proteins and importin beta recognizes the nuclear pore complex. This interaction is essential for the regulation of gene expression DNA replication and cell cycle progression. KPNA6 participates in coordinating these cellular processes by ensuring proteins enter the nucleus with timely precision.
Nuclear transport mechanisms involve KPNA6 prominently. It plays a significant role in the nucleocytoplasmic transport pathway also contributing to the regulation of the cell cycle.
