Supplementary information

KRT6B also known as Keratin 6B or CK6B is a type II cytoskeletal keratin. This protein has a molecular mass of approximately 60 kDa. It is mostly found in epithelial cells particularly in stratified squamous epithelia such as the epidermis hair follicles and oral mucosa. KRT6B participates in forming a structural network inside cells providing resilience to mechanical stress and contributing to the integrity and function of epithelial layers.
Biological function summary

KRT6B engages in cellular roles across various tissues. It is involved in the keratinocyte activation cycle and plays a part in wound healing processes. KRT6B forms a heterodimer complex with type I keratins like KRT16 and KRT17 stabilizing intermediate filaments that maintain cell structure. This assembly assists in skin health and influences cellular behaviors such as migration and differentiation especially during tissue repair.

Pathways

KRT6B integrates into signaling pathways like the Notch and Wnt pathways which regulate differentiation and proliferation in epithelial tissues. Within these pathways KRT6B interacts with proteins such as Notch receptors and components of the Wnt signaling cascade. Its association with these pathways highlights its role in cellular fate decisions and epidermal homeostasis reflecting its relevance to both normal and pathophysiological processes.

KRT6B links to conditions like pachyonychia congenita and psoriasis. Mutations in KRT6B can cause pachyonychia congenita where it affects nail and skin integrity. Additionally in psoriasis abnormal KRT6B expression relates to the rapid turnover of keratinocytes. Its interaction with keratin pairs like KRT16 and KRT17 during these disorders highlights its role in disease mechanisms and suggests potential as a therapeutic target.
