Supplementary information

KRT81 also known as Keratin 81 or Hair Keratin K2.9 is a type II keratin protein with a molecular mass of approximately 55 kDa. This protein is mechanical forming intermediate filaments that provide structural support in the cells. KRT81 is primarily found in hair follicles especially in the cortex of hair fibers where it plays a role in the formation and stabilization of hair structure. Its highly regulated expression is critical during hair development and growth.
Biological function summary

KRT81 contributes to the rigidity and integrity of hair by interacting within a keratin complex. This complex includes other keratin proteins that support the polymerization process needed for filament formation. The presence of KRT81 in non-dividing differentiating cells in the hair follicle indicates its important role during the hair cycle stages ensuring the mechanical strength needed for protective and sensory functions.

Pathways

Several biological processes depend on the structural role of KRT81. It is part of the keratinization pathway which is essential for hair formation impacting the assembly of hair alongside other keratin proteins such as KRT31 and KRT35. These proteins work together to create the strong keratin structure necessary for healthy hair development. KRT81 interacts within a network that influences hair shaft differentiation.

KRT81 mutations can lead to hair-related conditions. One such disorder is Monilethrix characterized by beaded hair and hair shaft abnormalities. In Monilethrix changes in KRT81 can disrupt the protein network impacting other keratins like KRT83 and leading to fragile hair. Understanding these connections helps in developing therapies aimed at treating hair-related disorders by targeting specific points in the keratin protein network.
