Immunofluorescent analysis of 4% PFA fixed, 0.1% Triton X-100 in PBS permeabilized A549 (Human lung carcinoma cell line) cells labeling L-Kynurenine Hydrolase with ab276053 at 1/60 dilution, 4°C overnight, folloed by Goat Anti-rabbit IgG Alexa Fluor 488-conjugated secondary antibody (green). Positive staining was localized to Cytoplasm. Counterstained with DAPI (blue). Cells were blocked with 10% serum prior incubation.

KYNU-HIS was immunoprecipitated using 0.5 mg of A549 (Human lung carcinoma cell line) whole cell lysate with 0.5 μL ab276053 and 15 μl of 50 % Protein G agarose. Western blot was performed from the immunoprecipitate using ab276053 at 1/500 dilution. Clean-Blot IP Detection Reagent (HRP), was used as secondary antibody at 1/1000 dilution.

Developed using the DAB staining technique.
Performed under reducing conditions.
