Polycomb group (PcG) protein that specifically recognizes and binds mono- and dimethyllysine residues on target proteins, therey acting as a 'reader' of a network of post-translational modifications. PcG proteins maintain the transcriptionally repressive state of genes: acts as a chromatin compaction factor by recognizing and binding mono- and dimethylated histone H1b/H1-4 at 'Lys-26' (H1bK26me1 and H1bK26me2) and histone H4 at 'Lys-20' (H4K20me1 and H4K20me2), leading to condense chromatin and repress transcription. Recognizes and binds p53/TP53 monomethylated at 'Lys-382', leading to repress p53/TP53-target genes. Also recognizes and binds RB1/RB monomethylated at 'Lys-860'. Participates in the ETV6-mediated repression. Probably plays a role in cell proliferation. Overexpression induces multinucleated cells, suggesting that it is required to accomplish normal mitosis.

L3MBTL1 also known as the MBT domain-containing protein L3MBTL1 is a protein that plays a role in chromatin remodeling. This protein has a molecular mass of approximately 83 kDa. It is expressed in various tissues including hematopoietic progenitor cells and the brain. L3MBTL1 interacts with DNA and histones participating in the modification of chromatin structure important for gene expression regulation.
L3MBTL1 functions as a transcriptional repressor involved in cell cycle regulation and differentiation. It acts by binding to methylated lysines on histone tails affecting chromatin compaction and accessibility. L3MBTL1 is part of the Polycomb group (PcG) protein complex which maintains genes in a repressed state ensuring proper cell fate determination.
L3MBTL1 is an important component in chromatin modification and transcriptional repression pathways. It participates in pathways such as the Polycomb Repressive Complex 1 (PRC1)-mediated gene silencing pathway. L3MBTL1 interacts with other Polycomb proteins such as BMI1 and RING1 to ensure effective repression of target genes.
L3MBTL1 has a strong connection to certain types of cancer and genetic syndromes. Alterations in L3MBTL1 expression or function can contribute to hematological malignancies like acute myeloid leukemia. It is also linked to developmental disorders due to its role in epigenetic regulation during development. L3MBTL1 may interact with proteins like EZH2 a related PcG protein influencing the progression of these diseases.
