Is a negative regulator of Notch target genes expression, required for RBPJ-mediated transcriptional repression (PubMed:29030483). It recruits KDM1A to Notch-responsive elements and promotes KDM1A-mediated H3K4me demethylation (PubMed:29030483). Involved in the regulation of ubiquitin-dependent degradation of a set of methylated non-histone proteins, including SOX2, DNMT1 and E2F1. It acts as an adapter recruiting the CRL4-DCAF5 E3 ubiquitin ligase complex to methylated target proteins (PubMed:29691401, PubMed:30442713). Required for normal maturation of myeloid progenitor cells (By similarity).

Activity summary

L3MBTL3 also known as Lethal(3) Malignant Brain Tumor-Like Protein 3 is a member of the polycomb group proteins which regulate gene expression by remodeling chromatin. It has a molecular weight of approximately 69 kDa. L3MBTL3 is widely expressed in various tissues with significant levels in the brain and hematopoietic cells. This protein plays a role in chromatin compaction through its ability to recognize and bind mono- and dimethylated lysine residues on histones. These activity patterns classify it as a histone reader influencing the accessibility of transcriptionally active regions.

Biological function summary

L3MBTL3 affects cellular processes by managing chromatin structure which in turn regulates gene expression. It participates in chromatin remodeling as part of large multi-protein complexes.
