Supplementary information

L3MBTL4 also known as L(3)mbt-like protein 4 belongs to the Polycomb group of proteins. It has a molecular mass of approximately 110 kDa. This protein mainly functions as a chromatin reader recognizing and binding to methylated histone tails. L3MBTL4 localizes to the nucleus where it is involved in chromatin remodeling. Expression of L3MBTL4 occurs in many tissues with higher levels seen in brain and testes suggesting a role in these areas.
Biological function summary

L3MBTL4 influences chromatin state and gene expression by interacting with other chromatin-associated proteins participating in the silencing of genes. This protein is a part of larger complexes involved in chromatin modification notably influencing transcriptional repression activities. It contributes to the regulation of various genes thereby impacting cell cycle progression and differentiation events within cells.

Pathways

L3MBTL4 participates in the regulation of key processes such as the cell cycle and epigenetic modification pathways. It interacts with proteins like RB and members of the PcG complex to modulate these pathways ensuring appropriate cell proliferation and differentiation. L3MBTL4 also plays a role in histone modification pathways further highlighting its importance in gene expression regulation through its interaction with other chromatin modulators.

L3MBTL4 has been associated with certain cancers and neurodevelopmental disorders. Dysregulation of L3MBTL4 expression or function can contribute to tumorigenesis particularly influencing pathways involving RB protein and other chromatin-related proteins. Additionally its misregulation might impact neurodevelopmental pathways potentially linking it to disorders such as intellectual disability where it may interact with proteins implicated in neuronal development.

Target data

Putative Polycomb group (PcG) protein. PcG proteins maintain the transcriptionally repressive state of genes, probably via a modification of chromatin, rendering it heritably changed in its expressibility (By similarity).
