Western blot - Anti-LAMA1 antibody [CL3087] (AB210954)
Lanes 1-3 : Merged signal (red and green). Green - ab210954 observed at 337 kDa. Red - loading control ab129002 observed at 124 kDa.
ab210954 Anti-LAMA1 antibody [CL3087] was shown to specifically react with LAMA1 in wild-type HeLa cells. Loss of signal was observed when knockout cell line ab264991 (knockout cell lysate ab258937) was used. Wild-type and LAMA1 knockout samples were subjected to SDS-PAGE. ab210954 and Anti-Vinculin antibody [EPR8185] (ab129002) were incubated overnight at 4°C at 1 in 1000 dilution and 1 in 20000 dilution respectively. Blots were developed with Goat anti-Rabbit IgG H&L (IRDye® 680RD) preadsorbed (ab216777) and Goat anti-Mouse IgG H&L (IRDye® 800CW) preadsorbed (ab216772) secondary antibodies at 1 in 20000 dilution for 1 hour at room temperature before imaging.

Biological function summary
This protein acts as a structural scaffold and cell adhesion mediator. LAMA1 integrates into the laminin-1 trimer complex composed of alpha beta and gamma chains supporting cellular attachment differentiation and migration. This assembly aids in tissue morphogenesis and repair processes. It creates an important interface between cells and the extracellular matrix influencing cellular behavior and maintaining tissue function.
Pathways
LAMA1 interacts with cellular signaling cascades such as the PI3K/AKT pathway and the TGF-beta pathway. These pathways play significant roles in cellular growth survival and differentiation. LAMA1's interaction with other proteins like integrins and heparan sulfate proteoglycans facilitates its role in these pathways impacting diverse processes like embryonic development and tissue remodeling.
