Activity summary

Laminin alpha 4 (LAMA4) also known as laminin-8 is a subunit of the laminin protein family which are essential components of the extracellular matrix. This protein has a molecular weight of approximately 210 kDa. LAMA4 mainly expresses in the vasculature including blood vessels and organs with high metabolic rates like the heart and skeletal muscles. It plays a significant role in maintaining the structural integrity of tissues by forming a network that supports cell adhesion and migration.

Biological function summary

Laminin alpha 4 interacts with other laminin subunits such as beta and gamma chains to form heterotrimeric laminin complexes. These laminins are important for cell differentiation proliferation and tissue repair. LAMA4 influences the basement membrane dynamics contributing to the biomechanical properties of cells. Through its binding to integrins and other cell surface receptors LAMA4 activates intracellular signaling cascades necessary for organogenesis and tissue regeneration.

Pathways

Laminin alpha 4 participates actively in the PI3K/AKT pathway important for cell survival and growth. It also plays a role in the MAPK signaling pathway which is important for cell differentiation and response to stress signals. Through these pathways LAMA4 interacts with various proteins including integrins and dystroglycan modulating essential cellular processes and maintaining cellular homeostasis.

Associated diseases and disorders

LAMA4's role becomes prominent in cancer progression where altered expression can influence tumor invasion and metastasis. Furthermore it connects to cardiovascular disorders especially in influencing angiogenesis and repair of ischemic tissues. In these contexts LAMA4 relates to proteins like integrin alpha6 and beta1 which facilitate aberrant cellular activities in disease processes. Understanding LAMA4's involvement offers potential pathways for therapeutic intervention in these diseases.
