Required for optimal lysosomal function (PubMed:21224396). Blocks EGF-stimulated EGFR intraluminal sorting and degradation. Conversely by binding with the phosphatidylinositol 4,5-bisphosphate, regulates its PIP5K1C interaction, inhibits HGS ubiquitination and relieves LAPTM4B inhibition of EGFR degradation (PubMed:25588945). Recruits SLC3A2 and SLC7A5 (the Leu transporter) to the lysosome, promoting entry of leucine and other essential amino acid (EAA) into the lysosome, stimulating activation of proton-transporting vacuolar (V)-ATPase protein pump (V-ATPase) and hence mTORC1 activation (PubMed:25998567). Plays a role as negative regulator of TGFB1 production in regulatory T cells (PubMed:26126825). Binds ceramide and facilitates its exit from late endosome in order to control cell death pathways (PubMed:26280656).

LAPTM4B also known as Lysosomal Protein Transmembrane 4 Beta is a membrane protein involved in endolysosomal trafficking. This protein has a molecular mass of around 27 kDa. LAPTM4B is highly expressed in liver kidney and placenta tissue with notable expression in several types of solid tumors. Its location in endosomes and lysosomes suggests a role in cellular cargo sorting and transport processes.
LAPTM4B is essential for regulating lysosomal function and intracellular signaling. This protein forms part of a complex with other lysosomal associated proteins and is involved in the regulation of lysosome positioning and signaling pathways. By modulating lysosome function LAPTM4B affects cellular processes like growth proliferation and apoptosis playing an important role in cell survival and homeostasis.
This protein participates in the PI3K/Akt and MAPK signaling pathways which are critical for cell growth and survival. LAPTM4B interacts with proteins such as EGFR (Epidermal Growth Factor Receptor) and mTOR (mammalian target of rapamycin) which facilitate its role in these pathways. Its involvement in these pathways contributes to its regulatory function in cell survival and metabolism.
Researchers link LAPTM4B to cancer and osteoporosis. Overexpression of LAPTM4B has been observed in various types of cancers including liver and breast cancer where it may contribute to tumor growth and metastasis. In osteoporosis LAPTM4B interacts with proteins such as Cathepsin K which are involved in bone resorption suggesting its potential role in bone metabolism and health.

Immunocytochemistry/ Immunofluorescence anaysis of A431 cells labeling LAPTM4B with ab242376 at 2-10 μg/mL. Cells were fixed with PFA and permeabilised with Triton X-100.
