Supplementary information

LARG also known as Leukemia Associated Rho Guanine Nucleotide Exchange Factor is a RhoGEF involved in the regulation of Rho GTPases. It functions mechanically by facilitating the exchange of GDP for GTP on Rho proteins thereby activating them. LARG has a molecular mass of approximately 173 kDa. It expresses in various tissues including the brain heart and kidney highlighting its potential involvement in diverse biological processes.
Biological function summary

LARG influences cytoskeletal dynamics and cell mobility. It associates with G-protein coupled receptor signaling complexes impacting cellular responses to external stimuli. This protein also has important roles in the regulation of cell adhesion migration and proliferation. Through interactions with other proteins such as PDZ-RhoGEF and p115-RhoGEF LARG influences the organization of the actin cytoskeleton and is involved in maintaining tissue architecture.

Pathways

LARG integrates into the RhoA signaling pathway and is an important player in the regulation of cellular structure and motility. It interacts with proteins involved in the Rho GTPase cycle such as RhoA and RhoC and modulates actin cytoskeletal arrangement. LARG also participates in the Wnt signaling pathway influencing gene expression and cell behavior. Through these interactions LARG contributes to regulating processes like cell morphology and division.

LARG has connections to certain cancers and cardiovascular disorders. It interacts with proteins such as Gα13 and RhoA which have implications in the progression of these diseases. In cancer alterations in LARG expression or function may lead to enhanced cell proliferation and metastasis. In cardiovascular disorders LARG-related pathways might affect cell contraction and architecture in vascular tissues contributing to pathological states. Understanding LARG's role in these contexts could help develop targeted therapies.

Target data

May play a role in the regulation of RhoA GTPase by guanine nucleotide-binding alpha-12 (GNA12) and alpha-13 (GNA13). Acts as guanine nucleotide exchange factor (GEF) for RhoA GTPase and may act as GTPase-activating protein (GAP) for GNA12 and GNA13.
