Western blot - Anti-LARP1 antibody [EPR28797-92] (AB317575)
Western blot : Anti-LARP1 antibody [EPR28797-92] ab317575 staining at 1/10000 dilution, shown in green; Mouse anti alpha Tubulin (ab7291) loading control staining at 1/20,000 dilution, shown in magenta. A band was observed at 150 kDa in Wild-type A549 cell lysates with no signal observed at this size in LARP1 knockout A549 cell line. To generate this image, samples were run on an SDS-PAGE gel then transferred onto a nitrocellulose membrane. Membranes were blocked in 100pc LICOR in TBS-0.1 % Tween® 20 (TBS-T) before incubation with primary antibodies overnight at 4 °C. Blots were washed four times in TBS-T, incubated with secondary antibodies for 1 h at room temperature, washed again four times then imaged. Secondary antibodies used were Goat anti-Rabbit 800CW & Goat anti-Mouse 680RD at 1/20,000 dilution.

Biological function summary
LARP1 acts as an important regulator in the translation of mRNA particularly within the mTOR (mechanistic target of rapamycin) signaling pathway. LARP1 functions as part of a molecular complex that includes mTOR and other translation-associated proteins. This involvement in the mTOR pathway links LARP1 to protein synthesis control which is important for cell growth proliferation and survival. The interaction between LARP1 and mTORC1 (mTOR complex 1) enhances the translational efficiency of certain mRNAs influencing cellular responses to nutritional and growth signals.
Pathways
LARP1 interacts significantly with the mTORC1 signaling pathway which is important in cell growth and metabolism regulation. Its activity connects to proteins like eukaryotic translation initiation factor 4E-binding protein 1 (4EBP1) that are important for controlling protein synthesis. The function of LARP1 in these pathways helps maintain cellular homeostasis especially under varying environmental conditions by modulating the translation of mRNAs that encode for ribosomal proteins and other components of the translational machinery.
