LAS1L also known as LAS1-like protein is a nucleolar protein with a molecular mass of approximately 80 kDa. It is expressed widely across human tissues with prominent presence in actively dividing cells. Mechanically LAS1L assists in the processing and maturation of pre-ribosomal RNA (rRNA) playing a critical role in ribosome biogenesis. This protein functions as part of a multi-protein complex ensuring precise assembly and function of ribosomes.
Biological function summary

LAS1L serves significant functions in the nucleolus within cell nucleuses. As a component of a larger complex called the pre-ribosome LAS1L coordinates with other proteins to facilitate the cleavage of pre-rRNA into smaller ribosomal RNA units. This activity is essential for producing functional ribosomal units which are important for protein synthesis. Its interaction with proteins such as GTPases and endonucleases is important for maintaining ribosome assembly fidelity.

Pathways

LAS1L operates within the ribosome biogenesis pathway closely interacting with proteins like PELP1 and NOL9. It plays a role in the mTOR signaling pathway impacting cell growth and proliferation. The protein also helps regulate downstream processes critical for the final assembly of ribosomal subunits integrating cellular nutrient signals with ribosome production.

LAS1L shows connections to various cancers and ribosomopathies. Alterations in LAS1L expression or function can lead to disrupted ribosomal biogenesis contributing to cancer pathogenesis due to uncontrolled cell division. Moreover LAS1L is linked with Diamond-Blackfan Anemia a disorder of ribosome production. Related proteins like NPM1 and RPL11 are involved in this condition highlighting the interplay between LAS1L and ribosome function-related diseases.

Target data

Required for the synthesis of the 60S ribosomal subunit and maturation of the 28S rRNA (PubMed : 20647540). Functions as a component of the Five Friends of Methylated CHTOP (5FMC) complex; the 5FMC complex is recruited to ZNF148 by methylated CHTOP, leading to desumoylation of ZNF148 and subsequent transactivation of ZNF148 target genes (PubMed : 22872859). Required for the efficient pre-rRNA processing at both ends of internal transcribed spacer 2 (ITS2) (PubMed : 22083961).
