LASS3 also known as Ceramide Synthase 3 or CERS3 is a protein that catalyzes the synthesis of ceramides essential components of lipid metabolism. It exhibits a molecular mass of approximately 45 kDa. LASS3 is expressed in various tissues but shows a higher expression in the skin indicating a potential role in dermatological functions. Furthermore LASS3 resides mainly in the endoplasmic reticulum where it executes its enzymatic functions efficiently.
Biological function summary

Ceramide synthesis by LASS3 contributes significantly to the maintenance of skin barrier functions and epidermal keratinization processes. LASS3 acts as part of the larger ceramide synthase family which forms ceramide as a central molecule in sphingolipid metabolism. Keratinocytes display a reliance on LASS3 activity to produce long-chain ceramides which are key in forming the lipid matrix of the skin.

Pathways

LASS3 participates in the sphingolipid metabolic pathway working alongside other ceramide synthases like LASS1 and LASS2. It contributes to the formation of ceramides involved in cell signaling and apoptosis pathways. LASS3's activity links to the Protein Kinase C signaling pathway which regulates cell proliferation and differentiation by interacting with PKC isoforms influencing downstream signaling events.

LASS3 plays a role in skin-related conditions such as ichthyosis and psoriasis. Ceramide synthase that catalyzes the transfer of the acyl chain from acyl-CoA to a sphingoid base, with high selectivity toward very- and ultra-long-chain fatty acyl-CoA (chain length greater than C22) (PubMed : 17977534, PubMed : 22038835, PubMed : 26887952). N-acylates sphinganine and sphingosine bases to form dihydroceramides and ceramides in de novo synthesis and salvage pathways, respectively (PubMed : 17977534, PubMed : 22038835, PubMed : 26887952).
