The LAYN protein also known as layilin plays a role in cell adhesion and signal transduction. It is an approximately 55 kDa protein expressed mainly in epithelial and immune cells including T-cells and natural killer cells. This protein sits on the cell surface and has a connection to the cytoskeleton through its cytoplasmic tail. Alternate expression may occur in both normal and cancerous tissues indicating a role in various physiological and pathological processes.
Biological function summary

LAYN facilitates interactions between cells and the extracellular matrix. It acts as a receptor for hyaluronic acid contributing to cell migration and tissue remodeling. While it is not part of any specific complex layilin may interact with other extracellular matrix proteins to promote cellular activities. This interaction influences immune responses by modulating immune cell activation and homing.

Pathways

LAYN is involved in the hyaluronan-mediated motility pathway where it binds hyaluronic acid to regulate cell movement. Additionally the protein takes part in the immune response pathway by influencing immune cell behavior. LAYN interacts with proteins like moesin which helps connect the cell membrane to the actin cytoskeleton driving cellular changes during these processes.

LAYN has a connection to cancer progression and autoimmune diseases. It plays a role in the tumor microenvironment affecting tumor cell motility and can influence metastasis. In autoimmune disorders LAYN's role in modulating immune cell activity can trigger aberrant immune responses. The protein interacts with various components of the immune system linking its expression to both health and disease states.
