The target proteins LC3A and LC3B also known as MAP1LC3A and MAP1LC3B are part of the microtubule-associated protein 1 light chain 3 family. Each of these proteins has a molecular mass of approximately 17 kDa. They are present in various tissues throughout the body including the brain liver and heart. LC3A/B proteins are expressed in both the cytoplasm and autophagic vesicles where they play important roles in autophagy mechanisms.
Biological function summary

LC3A/B proteins serve as central markers and mediators in the autophagy process where they are involved in the formation of autophagosomes. These proteins exist as part of the complex machinery that includes other autophagy proteins like ATG7 and ATG3. LC3A/B facilitate the elongation and closure of the phagophore membrane forming complete autophagosomes that encapsulate cellular debris and damaged organelles for degradation.

Pathways

LC3A/B proteins are integral to the autophagy pathway which maintains cellular homeostasis by recycling cellular components. Autophagy is linked to the mTOR signaling pathway where mTOR acts as an important regulator. LC3A/B interacts with the ubiquitin-proteasome system and proteins like p62/SQSTM1 that recognize ubiquitinated cargo directing them toward degradation in autophagosomes.

LC3A/B proteins are implicated in conditions like neurodegenerative diseases including Parkinson's disease where autophagy dysfunction plays an important role. Aberrant regulation of LC3A/B and connected proteins like alpha-synuclein contribute to the pathology of these diseases. LC3A/B is also associated with cancer where alterations in autophagic activity often lead to tumor progression or suppression depending on the context and tumor type.

Target data

Ubiquitin-like modifier involved in formation of autophagosomal vacuoles (autophagosomes) (PubMed : 20418806, PubMed : 23209295, PubMed : 28017329). Plays a role in mitophagy which contributes to regulate mitochondrial quantity and quality by eliminating the mitochondria to a basal level to fulfill cellular energy requirements and preventing excess ROS production (PubMed : 23209295, PubMed : 28017329). In response to cellular stress and upon mitochondria fission, binds C-18 ceramides and anchors autophagolysosomes to outer mitochondrial membranes to eliminate damaged mitochondria (PubMed : 22922758). While LC3s are involved in elongation of the phagophore membrane, the GABARAP/GATE-16 subfamily is essential for a later stage in autophagosome maturation (PubMed : 20418806, PubMed : 23209295, PubMed : 28017329). Promotes primary ciliogenesis by removing OFD1 from centriolar satellites via the autophagic pathway (PubMed : 24089205). Through its interaction with the reticulophagy receptor TEX264, participates in the remodeling of subdomains of the endoplasmic reticulum into autophagosomes upon nutrient stress, which then fuse with lysosomes for endoplasmic reticulum turnover (PubMed : 31006537, PubMed : 31006538). Upon nutrient stress, directly recruits cofactor JMY to the phagophore membrane surfaces and promotes JMY's actin nucleation activity and autophagosome biogenesis during autophagy (PubMed : 30420355).
