The protein LC3C also known as microtubule-associated protein 1 light chain 3 gamma is part of the ATG8 family and is critical in the autophagy pathway. LC3C with a molecular mass of approximately 14.7 kDa participates in the formation of autophagosomes. This protein predominantly localizes in the cytoplasm and transitions to autophagosome membranes during autophagy. LC3C expression occurs in multiple tissues including the brain and liver indicating its broad role in cellular function.
Biological function summary

Microtubule-associated protein 1 light chain 3 gamma acts as an important component in the autophagy process. It is part of a complex that involves other homologs of the LC3 protein family. LC3C interacts directly with phosphatidylethanolamine on autophagosome membranes contributing significantly to the expansion and sealing of these structures. This action is critical for cellular homeostasis and survival during nutrient deprivation.

Pathways

The protein LC3C is an integral part of the macroautophagy pathway. It associates with proteins such as p62 (SQSTM1) which delivers ubiquitinated cargo to autophagosomes. LC3C also interacts with Beclin-1 a protein involved in the initiation of autophagy. These interactions place LC3C within the autophagic machinery essential for lysosomal degradation and recycling of cellular components facilitating proper cellular function.

Research indicates that microtubule-associated protein 1 light chain 3 gamma relates to cancer and neurodegenerative disorders. In cancer altered LC3C levels can affect tumor cell survival and adaptation to stress associating with proteins such as PINK1 which regulates mitochondrial autophagy. In neurodegenerative diseases improper autophagy due to dysfunctional LC3C may contribute to the accumulation of damaged proteins leading to conditions like Parkinson's disease where LC3C interacts with parkin an important player in targeting dysfunctional mitochondria for removal through mitophagy.
