LCMT1 also known as leucine carboxyl methyltransferase 1 is an enzyme with a mass of approximately 25 kDa. This enzyme plays an important role in the methylation of the C-terminal leucine residues of proteins specifically targeting protein phosphatase 2A (PP2A). It methylates the PP2A catalytic subunit affecting its activity and interactions. LCMT1 expression occurs widely in tissues with high expression noted in the brain and testis.
Biological function summary

This enzyme contributes to the dynamic regulation of protein phosphatase activity within cells. LCMT1 operates as part of a complex with PP2A and influences the formation of PP2A holoenzymes. By methylating the catalytic subunit of PP2A LCMT1 impacts the holoenzyme's substrate specificity and enzymatic activity contributing to cellular signaling and homeostasis.

Pathways

LCMT1 activity integrates into the regulation of various signaling cascades notably the MAPK and AKT pathways. These pathways are important for cell growth survival and differentiation. LCMT1 influences these pathways by modulating PP2A activity which can dephosphorylate key signaling molecules such as ERK and AKT. LCMT1 works in concert with other proteins like PME-1 that demethylate PP2A to fine-tune this regulation.

LCMT1 has connections to neurodegenerative diseases and certain cancers. In neurodegenerative diseases such as Alzheimer's disease altered LCMT1 activity can lead to dysregulation of PP2A which affects tau phosphorylation and accumulation. Moreover in cancer such as hepatocellular carcinoma LCMT1's regulation of PP2A can influence cellular proliferation and apoptosis. It interacts functionally with proteins like alpha4 which is associated with PP2A and involved in their pathogenesis.
