The LDHD/DLD protein also known as D-lactate dehydrogenase and capable of forming a complex with DLD (dihydrolipoamide dehydrogenase) has a molecular mass approximately 60 kDa. It mainly functions as an enzyme responsible for catalyzing the conversion of D-lactate to pyruvate in the mitochondria. Expression of LDHD is primarily observed in metabolic tissues such as liver kidney and heart. This protein plays an important role in energy production and metabolism by contributing to cellular redox balance.
Biological function summary

LDHD/DLD has an essential role in the mitochondrial energy metabolism through its involvement in the D-lactate metabolism. While LDHD/DLD itself is not formally a part of the pyruvate dehydrogenase complex DLD present in the complex shares the enzymatic responsibility for electron transfer needed for efficient energy conversion. The enzymatic action of LDHD in concert with DLD maintains cellular energy balance and supports effective metabolic processes.

Pathways

Research has shown connections between LDHD/DLD activity and the pyruvate metabolism and glycolysis pathways. In these pathways LDHD/DLD helps in channeling D-lactate derived electrons into the electron transport chain facilitating energy production in the form of ATP. The interaction between LDHD and proteins like pyruvate dehydrogenase ties them to those pathways ensuring proper cellular respiration and metabolic regulation within cells that demand high energy.

LDHD/DLD has been linked to issues involving lactic acidosis and some metabolic syndromes. A dysfunction in the enzymatic activity of LDHD/DLD could lead to the accumulation of D-lactate contributing to lactic acidosis which disrupts normal pH balance. Disorders in metabolic pathways involving DLD and related proteins can also manifest in altered energy metabolism linking LDHD/DLD to conditions like mitochondrial disorders and some forms of diabetes.
