Supplementary information

LETM1 short for Leucine Zipper and EF-Hand Containing Transmembrane Protein 1 plays an important role in cellular ion regulation. This protein with a mass of approximately 81 kDa is mainly located in the inner mitochondrial membrane. LETM1 acts as a mitochondrial calcium and potassium exchanger. It facilitates the transport of calcium out of the mitochondria balancing mitochondrial ion homeostasis. Expression of LETM1 occurs widely across different tissue types indicating its importance in basic cellular function.
Biological function summary

LETM1 contributes to maintaining mitochondrial function and integrity. It comprises part of a complex that includes other mitochondrial membrane proteins. This complex ensures sufficient ion gradients and energy production within the cell. Its role in calcium ion homeostasis aids in regulating mitochondrial membrane potential and ATP synthesis. Efficient operation of this mechanism is essential for cell survival and function.

Pathways

LETM1 integrates into metabolic pathways where it supports mitochondrial bioenergetics and oxidative phosphorylation. It interacts with the OXPHOS system during energy production. LETM1's activity also affects the cellular apoptosis pathway influenced by its regulation of calcium levels. Related proteins like MICU1 and MCU participate in these pathways contributing to the regulation of calcium uptake into mitochondria and further connecting LETM1’s role to broader cellular processes.

LETM1 is linked to Wolf-Hirschhorn syndrome a genetic disorder characterized by developmental delays and seizures. Mutations or deletions affecting LETM1 contribute significantly to the syndrome's phenotype. Additionally LETM1 dysregulation associates with certain types of cancer where aberrant calcium signaling may promote tumor progression. The connection between LETM1 and proteins like Cyclophilin D involved in mitochondrial integrity highlights its influence in disease mechanisms.

Target data

Plays an important role in maintenance of mitochondrial morphology and in mediating either calcium or potassium/proton antiport (PubMed : 18628306, PubMed : 19797662, PubMed : 24344246, PubMed : 24898248, PubMed : 29123128, PubMed : 32139798, PubMed : 36055214, PubMed : 36321428). Mediates proton-dependent calcium efflux from mitochondrion (PubMed : 19797662, PubMed : 24344246, PubMed : 29123128). Also functions as an electroneutral mitochondrial proton/potassium exchanger (PubMed : 24898248, PubMed : 36055214, PubMed : 36321428). Crucial for the maintenance of mitochondrial tubular networks and for the assembly of the supercomplexes of the respiratory chain (PubMed : 18628306, PubMed : 36055214). Required for the maintenance of the tubular shape and cristae organization (PubMed : 18628306, PubMed : 32139798).
