Lgi4 also known as leucine-rich glioma inactivated 4 or Lgi-4 is a member of the Lgi protein family. This protein has an approximate mass of 120 kDa. Lgi4 plays an important structural role contributing to cell-cell communication in the nervous system. It is expressed in the peripheral nervous tissue specifically in Schwann cells which are part of the myelination process of peripheral nerves.
Biological function summary

Lgi4 participates in Schwann cell proliferation and differentiation impacting the myelination of peripheral nerves. As part of the non-covalent complex involving other Lgi family members it ensures the structural integrity and function of the myelin sheath. It regulates the interactions between neurons and glial cells promoting proper nerve function and development.

Pathways

Lgi4 interacts with the Nogo and Notch signaling pathways which are important for nerve growth and regeneration. In these pathways Lgi4 establishes interactions with Nogo receptor proteins and Notch receptors modulating signal transmission and myelin formation. These pathways play significant roles in neural development and repairing processes.

Lgi4 has associations with Charcot-Marie-Tooth disease and peripheral neuropathy. Mutations or dysregulation of Lgi4 can lead to defective myelination contributing to the pathology of these conditions. Its connection with neuregulins and myelin protein zero is important as these proteins are also involved in the myelination defects observed in these disorders. This highlights Lgi4's role in maintaining neural health and potential therapeutic interest.
