LHX3 also known as LIM homeobox 3 protein is a transcription factor involved in the development and differentiation of specific tissues in the human body. It is expressed mainly in the pituitary gland forebrain and spinal cord. The molecular weight of LHX3 is approximately 45 kDa. This transcription factor contains two LIM domains and one homeodomain which are significant for its DNA-binding capability. Through these domains LHX3 regulates the expression of target genes critical for tissue-specific development.
Biological function summary

LHX3 is essential for the proper formation of the pituitary gland and motor neurons. It acts as part of a complex that includes other LIM homeodomain proteins to modulate transcription of genes responsible for lineage specification. LHX3 functions by binding to DNA sequences and affecting the transcriptional activation or repression of gene targets influencing processes like hormone biosynthesis in the pituitary. This regulation ensures appropriate cellular responses during development and differentiation.

Recognizes and binds to the consensus sequence motif 5'-AATTAATTA-3' in the regulatory elements of target genes, such as glycoprotein hormones alpha chain CGA and visual system homeobox CHX10, positively modulating transcription; transcription can be co-activated by LDB2. Synergistically enhances transcription from the prolactin promoter in cooperation with POU1F1/Pit-1 (By similarity). Required for the establishment of the specialized cells of the pituitary gland and the nervous system (PubMed : 21149718). Involved in the development of interneurons and motor neurons in cooperation with LDB1 and ISL1 (By similarity).
