LHX3 is essential for the proper formation of the pituitary gland and motor neurons. It acts as part of a complex that includes other LIM homeodomain proteins to modulate transcription of genes responsible for lineage specification. LHX3 functions by binding to DNA sequences and affecting the transcriptional activation or repression of gene targets influencing processes like hormone biosynthesis in the pituitary. This regulation ensures appropriate cellular responses during development and differentiation.
LHX3 also known as LIM homeobox 3 protein is a transcription factor involved in the development and differentiation of specific tissues in the human body. It is expressed mainly in the pituitary gland forebrain and spinal cord. The molecular weight of LHX3 is approximately 45 kDa. This transcription factor contains two LIM domains and one homeodomain which are significant for its DNA-binding capability. Through these domains LHX3 regulates the expression of target genes critical for tissue-specific development.
LHX3 interacts within the Wnt and Notch signaling pathways which are pivotal in developmental processes. In the Wnt pathway it modulates gene expression that governs cell proliferation and differentiation. The Notch pathway involvement highlights its role in neural differentiation. Through these pathways LHX3 works closely with proteins like β-catenin in the Wnt pathway impacting how cells communicate and respond to developmental cues.
LHX3 mutations are associated with Combined Pituitary Hormone Deficiency (CPHD) and pituitary hormone synthesis issues resulting in growth and developmental disorders. LHX3 defects can interact with other proteins like PIT1 influencing the clinical presentation of CPHD. The understanding of LHX3's role in these pathways and its interaction with proteins offers insight into therapeutic approaches to mitigate these disorders.

ab124697 at 1/10 dilution, staining LHX3/LIM in permeabilized Jurkat cells by intracellular flow cytometry (red), compared with a negative control Rabbit IgG antibody (green).
We have systematically measured KD (the equilibrium dissociation constant between the antibody and its antigen), of more than 840 recombinant antibodies to assess not only their individual KD values but also to see the average affinity of antibody.
Based on the comparison with published literature values for mouse monoclonal antibodies, Recombinant antibodies appear to be on average 1-2 order of magnitude higher affinity.
