LIN54 also known as protein LINT-54 is a component of the DREAM complex involved in cell cycle regulation. This protein has a molecular weight of around 70 kDa and is expressed in various tissues throughout the body including the brain and lungs. LIN54 contains specific DNA-binding domains that enable it to interact directly with DNA allowing it to regulate the transcription of target genes. By binding to specific DNA sequences this protein helps control key biological processes necessary for cell proliferation.
Biological function summary

In coordination with other proteins LIN54 plays an important role in the regulation of the cell cycle. It forms part of the multiprotein DREAM complex a regulatory unit that shifts cells from proliferation to a quiescent state by repressing cell cycle-dependent genes. During the cell cycle LIN54 supports transitions between different phases by regulating gene expression connected to cell cycle arrest and activation ensuring cells divide correctly and avoid errors that could lead to pathological states.

Pathways

Multiple cell cycle pathways involve the participation of LIN54 influencing cellular transitions and growth signals. LIN54 is significantly associated with the E2F transcription factor pathway which regulates genes required for cell cycle progression. Another pathway that LIN54 plays a part in is the p53 signaling pathway where it interacts with proteins like RB1. These pathways help maintain cellular homeostasis and prevent unregulated growth that may result in various diseases.

LIN54 shows connections to cancer and neurodegenerative diseases. Aberrant expression or regulation of LIN54 could lead to uncontrollable cell proliferation seen in cancers. One of its associations in cancer involves the interaction with RB1 commonly mutated in various cancers. Additionally dysregulation in the DREAM complex including LIN54 might relate to neurodegenerative diseases like Alzheimer's where cell cycle control in neurons is disrupted. In G0 phase, the complex binds to more than 800 promoters and is required for repression of E2F target genes (PubMed : 17531812, PubMed : 17671431).
