Immunofluorescent analysis of 4% Paraformaldehyde-fixed, 0.1% TritonX-100 permeabilized HEK-293T (human embryonic kidney epithelial cell) transfected with a LINE-1 ORF2p expression vector containing a myc tag. cells labelling LINE-1 ORF2p with ab263071 at 1/2000 (0.506 ug/ml) dilution, followed by ab150081 Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) preadsorbed antibody at 1/1000 dilution (Green).

LINE-1 ORF2p also known as L1 ORF2 protein is a critical component in the process of retrotransposition. This protein approximately 150 kDa in mass is encoded by the Long Interspersed Nuclear Element-1 (LINE-1) a type of non-long terminal repeat (non-LTR) retrotransposon. LINE-1 ORF2p contains reverse transcriptase and endonuclease activities which are necessary for inserting copies of LINE-1 DNA back into the genome. This protein is mainly expressed in germ cells and early embryonic tissues although its presence is noted in various somatic tissues in certain contexts.
Biological function summary

LINE-1 ORF2p facilitates the mobilization and insertion of LINE-1 elements into new genomic locations an activity integral to maintaining genome plasticity. It is not generally classified as part of a larger protein complex but functions closely with another protein LINE-1 ORF1p. ORF2p's reverse transcriptase domain synthesizes DNA from RNA allowing the LINE-1 element to integrate into the host genome. This action implicates it in genomic rearrangements that can have profound effects on gene expression and genome stability.

Pathways

LINE-1 ORF2p plays a substantial role in genomic and epigenomic pathways particularly those involving DNA synthesis and repair. It interacts with cellular proteins such as DNA repair machinery that modulate its insertion capacity and activity. This protein influences the development and function of cellular pathways through its impact on genomic architecture affecting neighboring gene regulation. The interaction with proteins like the Apurinic/apyrimidinic endonuclease (APE1) is significant as they can regulate LINE-1 insertion events.

LINE-1 ORF2p is associated with cancer and neurodegenerative diseases due to its role in genomic instability. Uncontrolled activity of LINE-1 can lead to mutagenic insertions in oncogenes or tumor suppressor genes contributing to tumorigenesis. Additionally in neurodegenerative diseases inappropriate activation of LINE-1 ORF2p may induce neuronal damage through genomic disarray. Related proteins include PIWI-interacting RNAs (piRNAs) which usually silence LINE-1 activity to prevent genomic chaos.

Has reverse transcriptase activity required for target-primed reverse transcription of the LINE-1 element mRNA, a crucial step in LINE-1 retrotransposition (PubMed : 38096901, PubMed : 38096902, PubMed : 7516468, PubMed : 9140393). Selectively binds and reversely transcribes RNA with a poly(A) tail consisting of at least 20 adenosines (PubMed : 38096901). Also has endonuclease activity that allows the introduction of nicks in the chromosomal target DNA (PubMed : 17626046, PubMed : 34554261, PubMed : 38096901, PubMed : 38096902, PubMed : 8945517). Cleaves DNA in AT-rich regions between a 5' stretch of purines and a 3' stretch of pyrimidines, corresponding to the sites of LINE-1 integration in the genome (PubMed : 8945517). Conformational properties of the target DNA sequence rather than specific nucleotides are key determinants of the ORF2p capacity for sequence-specific DNA recognition (PubMed : 17626046, PubMed : 34554261). Unlike related endonucleases, does not bend the DNA helix but causes compression near the cleavage site (PubMed : 34554261).
