LNK also known as SH2B3 is an adaptor protein about 70 kDa in mass. LNK contains an SH2 domain and is expressed mainly in hematopoietic cells including stem and progenitor cells. This protein inhibits signaling pathways by binding with receptors and blocking further transduction. It works by negatively regulating signals mediated by cytokine receptors and receptor tyrosine kinases.
Biological function summary

The LNK protein functions as a negative regulator in the hematopoietic system helping to control cell proliferation and immune response. LNK interacts with other signaling proteins as part of regulatory complexes often utilizing its SH2 domain to bind phosphorylated tyrosines. By doing so it inhibits the activity of various kinases involved in cell division ensuring proper signaling and growth control. This role is important to maintaining cellular homeostasis and preventing excessive cell proliferation.

Pathways

The LNK protein plays significant roles in the JAK-STAT and PI3K-AKT signaling pathways. In these pathways LNK interacts with signaling molecules like JAK2 and STAT5 to limit excessive signaling. This interaction is known to affect cellular responses to growth factors and cytokines impacting processes such as cell survival and proliferation. Through its activity in these pathways LNK helps ensure appropriate cellular responses to external stimuli.

LNK has connections with conditions like myeloproliferative neoplasms and hypertension. Mutations or aberrant expression of LNK can lead to dysregulated signaling that contributes to these diseases. In myeloproliferative neoplasms altered LNK function can lead to uncontrolled cell proliferation through interactions with proteins such as JAK2. In hypertension its role in regulating vascular tone and inflammation highlights its potential involvement in the pathogenesis of the disease.
