LNX2 also known as Ligand of Numb-protein X 2 is an E3 ubiquitin-protein ligase that plays a central role in controlling protein degradation. The molecular mass of LNX2 is approximately 73 kDa. This protein commonly appears in tissues such as the brain and liver. LNX2 contains several PDZ domains which facilitate protein-protein interactions and it is involved in the ubiquitination pathway marking proteins for degradation.
LNX2 interacts with other proteins to regulate cellular signaling processes. LNX2 often forms part of a larger protein complex that includes Numb a protein involved in cell fate determination. Through these interactions LNX2 modulates important cellular functions like endocytosis and cell signaling important for maintaining cellular homeostasis. LNX2's role in interacting with several proteins indicates its importance in cellular regulatory networks.
LNX2 significantly contributes to the Notch signaling pathway which regulates cell differentiation and proliferation. LNX2 interacts with the Numb protein within this pathway influencing the outcome of signal transduction. Additionally LNX2 has roles within the Wnt signaling pathway another critical pathway in cell fate decisions and embryonic development. These pathways highlight LNX2's involvement in regulating key cellular processes in developmental biology.
LNX2 has associations with cancers particularly in the brain and liver where its expression is prominent. Alterations in LNX2 expression or function may disrupt the balance of cellular signaling pathways and contribute to tumorigenesis. Additionally LNX2 has connections with neurological disorders potentially due to its regulation of Numb and subsequent effects on neurodevelopmental pathways. LNX2's involvement in these diseases underlines its potential as a therapeutic target.

Immunofluorescent analysis of A-431 cells fixed using 4% formaldehyde (reconstituted in 1X PBS) for 10 min at room temperature and permeabilized using 0.1 % Triton X-100 in PBS for 15 min at room temperature labeling LNX2 with ab313433 at 1 ?g/mL followed by Goat anti-Rabbit IgG (H+L) Secondary Antibody, Alexa Fluor 488 conjugate at 1/2500 dilution. Panel a) shows representative cells that were stained for detection and localization of LNX2 protein (green), Panel b) is stained for nuclei (blue) using DAPI at 1/50 dilution. Panel c) represents cytoskeletal F-actin staining using Alexa Fluor 594 Phalloidin at 1/200 dilution. Panel d) is a composite image of Panels a, b and c clearly demonstrating cytoplasmic localization of LNX2. Panel e) represents control cells with no primary antibody to assess background.
