Function

Component of the Wnt-Fzd-LRP5-LRP6 complex that triggers beta-catenin signaling through inducing aggregation of receptor-ligand complexes into ribosome-sized signalosomes. Cell-surface coreceptor of Wnt/beta-catenin signaling, which plays a pivotal role in bone formation. The Wnt-induced Fzd/LRP6 coreceptor complex recruits DVL1 polymers to the plasma membrane which, in turn, recruits the AXIN1/GSK3B-complex to the cell surface promoting the formation of signalosomes and inhibiting AXIN1/GSK3-mediated phosphorylation and destruction of beta-catenin. Required for posterior patterning of the epiblast during gastrulation (By similarity).

    Lane 1: Wild-type HAP1 cell lysate (20 μg)
    Lane 2: LRP6 knockout HAP1 cell lysate (20 μg)
    Lane 3: HeLa cell lysate (20 μg)

    Lanes 1 - 4: Merged signal (red and green). Green - ab134146 observed at 220 kDa. Red - loading control, ab18058, observed at 124 kDa.

    ab134146 was shown to recognize LRP6 when LRP6 knockout samples were used, along with additional cross-reactive bands. Wild-type and LRP6 knockout samples were subjected to SDS-PAGE. ab134146 and ab18058 (loading control to Vinculin) were diluted 1/10000 and 1/1000 respectively, and incubated overnight at 4°C. Blots were developed with Goat anti-Rabbit IgG H&L (IRDye® 800CW) preadsorbed (Goat anti-Rabbit IgG H&L (IRDye® 800CW) preadsorbed ab216773) and Goat anti-Mouse IgG H&L (IRDye® 680RD) preadsorbed (Goat anti-Mouse IgG H&L (IRDye® 680RD) preadsorbed ab216776) secondary antibodies at 1/10000 dilution for 1 hour at room temperature before imaging.
