Function

Auxiliary protein of the large-conductance, voltage and calcium-activated potassium channel (BK alpha). Modulates gating properties by producing a marked shift in the BK channel's voltage dependence of activation in the hyperpolarizing direction, and in the absence of calcium.

Activity summary

LRRC55 also known as Leucine Rich Repeat Containing 55 is a protein encoded by the LRRC55 gene. Mechanically LRRC55 functions as an auxiliary subunit modulating large-conductance calcium-activated potassium (BK) channels which are key players in maintaining cellular ionic homeostasis. This protein has an approximate mass of 37 kDa. LRRC55 is expressed across various tissues but it has a significant presence in the brain and to lesser extents in other tissues such as the testis and kidney.

Biological function summary

The LRRC55 protein contributes to fine-tuning the physiological properties of BK channels. It is not just a single operator but part of a complex involving these channels. BK channels are vital for regulating neuronal excitability muscle contraction and other fundamental cellular processes. Through modulating these channels LRRC55 helps in sculpting the kinetics and response to calcium therefore impacting the overall behavior of the cells where it is expressed.

Pathways

The role of LRRC55 in modulating BK channels integrates it into critical pathways such as the smooth muscle contraction and neuronal signal transduction pathways. These pathways involve calcium regulation where the BK channels play a pivotal role. LRRC55 relates closely to other proteins like KCNMA1 the main pore-forming subunit of BK channels thereby contributing to the functional properties of these channels in signaling pathways.

Associated diseases and disorders

LRRC55 has potential links with neurological disorders such as epilepsy due to its critical role in neuronal excitability and signaling. Disruption or dysregulation of BK channels where LRRC55 is an interacting partner may lead to altered synaptic activity and increased susceptibility to such disorders. Additionally aberrations in smooth muscle function linked to LRRC55 impact may hint toward a role in hypertension given the importance of BK channels in regulating vascular tone.
