Lipid droplet-associated protein that maintains the balance between lipogenesis and lipolysis and also regulates fatty acid oxidation in oxidative tissues. Recruits mitochondria to the surface of lipid droplets and is involved in lipid droplet homeostasis by regulating both the storage of fatty acids in the form of triglycerides and the release of fatty acids for mitochondrial fatty acid oxidation. In lipid droplet triacylglycerol hydrolysis, plays a role as a scaffolding protein for three major key lipolytic players: ABHD5, PNPLA2 and LIPE. Reduces the triacylglycerol hydrolase activity of PNPLA2 by recruiting and sequestering PNPLA2 to lipid droplets. Phosphorylation by PKA enables lipolysis probably by promoting release of ABHD5 from the perilipin scaffold and by facilitating interaction of ABHD5 with PNPLA2. Also increases lipolysis through interaction with LIPE and upon PKA-mediated phosphorylation of LIPE (By similarity).

LSDP5 also known as PLIN5 or Perilipin 5 is a protein that plays an important role in lipid droplet formation and regulation. Its molecular mass is approximately 52 kDa. This protein is mainly expressed in tissues with high lipid metabolism such as muscle and liver. LSDP5 localizes to the surface of lipid droplets where it regulates triglyceride storage and release connecting to various metabolic processes.
The protein contributes to the organization of lipid droplets and regulation of lipid storage in the cell. It is not typically part of a large protein complex. By modulating lipid droplet formation LSDP5 controls the flow of fatty acids in and out of these storage organelles. This action influences energy balance in cells and aids adaptation to metabolic changes supporting both lipid storage and mobilization as needed.
The protein influences energy homeostasis and lipid metabolism pathways notably the peroxisome proliferator-activated receptor (PPAR) pathway. It works in association with other lipid-related proteins including perilipin 1 and adipose triglyceride lipase coordinating lipid storage and hydrolysis. These interactions help regulate the balance between storage and use of lipids impacting mitochondrial energy production.
LSDP5 is connected to metabolic conditions like obesity and type 2 diabetes. Aberrations in its expression or function may lead to dysregulated lipid storage contributing to these metabolic disorders. It interacts with proteins such as PGC-1α which is involved in managing mitochondrial biogenesis and energy expenditure linking it to disease states characterized by impaired energy metabolism.
