LSM11 also known as U7 snRNP protein homolog is an important component in the spliceosomal snRNP complex specifically associated with U7 snRNP. LSM11 consists of a molecular mass of approximately 56 kDa. This protein expresses primarily in the nucleus where it plays a significant role in the processing of histone pre-mRNA. The mechanical action of LSM11 involves binding to RNA and facilitating the assembly of snRNP complexes which are essential in the maturation of histone transcripts.
Biological function summary

LSM11 acts within the U7 snRNP complex interacting with other snRNP proteins to ensure proper pre-mRNA cleavage and polyadenylation inhibition. This makes LSM11 essential for the correct formation of mature histone mRNA as histones require maturation without polyadenylation. The lack of poly-A tails in histone mRNA is vital for its role during DNA synthesis linking LSM11 directly to the cell cycle's intricacies. Its function is tightly regulated with multiple proteins interacting with LSM11 to modulate its activity within the histone gene expression pathway.

Pathways

LSM11 functionally integrates into the histone pre-mRNA processing pathway. This pathway critically contributes to cell cycle regulation particularly during DNA replication. LSM11 interacts with other key proteins such as FLASH and the U7 snRNA facilitating precise mRNA processing. Additionally LSM11 forms important interactions within the spliceosomal complex supporting the proper execution of splicing-related tasks necessary for cellular proliferation.

LSM11 has links to cell cycle-related abnormalities such as certain cancers where histone mRNA maturation is disrupted. Aberrant expression or functional disruption of LSM11 can lead to faulty histone mRNA processing contributing to tumorigenesis. The protein's association with other factors like FUS which also plays a role in RNA binding and processing highlights the complex interactions required to maintain normal cellular functions. Defects in LSM11 alongside disruptions in related proteins can manifest in various cancers where cell cycle regulation is impaired.

Target data

Component of the U7 snRNP complex that is involved in the histone 3'-end pre-mRNA processing (PubMed : 11574479, PubMed : 16914750, PubMed : 33230297). Increases U7 snRNA levels but not histone 3'-end pre-mRNA processing activity, when overexpressed (PubMed : 11574479, PubMed : 16914750). Required for cell cycle progression from G1 to S phases (By similarity). Binds specifically to the Sm-binding site of U7 snRNA (PubMed : 11574479, PubMed : 16914750).
