Plays a role in pre-mRNA splicing as component of the U4/U6-U5 tri-snRNP complex that is involved in spliceosome assembly, and as component of the precatalytic spliceosome (spliceosome B complex) (PubMed:28781166). The heptameric LSM2-8 complex binds specifically to the 3'-terminal U-tract of U6 snRNA (PubMed:10523320).

Activity summary

LSM4 also known as Like-Sm protein 4 is a component of the LSM protein family with a molecular mass of approximately 12 kDa. It plays a mechanical role in the formation of the LSM complex which is similar to the Sm proteins involved in RNA processing. Expression of LSM4 occurs in various tissues including the brain and immune cells indicating its diverse functional significance across different biological systems.

Biological function summary

Like-Sm protein 4 plays an important role in RNA processing particularly in mRNA splicing and degradation. It forms part of the LSm1-7 complex which associates with the U6 snRNA to regulate the splicing mechanisms necessary for proper gene expression. By ensuring accurate mRNA splicing LSM4 contributes to the maintenance of cellular function and homeostasis.

Pathways

LSM4's involvement in RNA splicing positions it within the spliceosome pathway essential for processing pre-mRNA into mature mRNA. Through this pathway it collaborates with other LSm proteins and the DEAD-box helicase DDX41. LSM4 also has a role in the RNA decay pathway where it interacts with the XRN1 protein facilitating the turnover of mRNA to control gene expression levels in cells.

Associated diseases and disorders

Errors in LSM4 function may lead to neurological disorders such as spinal muscular atrophy. This association occurs through its interaction with the SMN protein where disruptions in RNA splicing affect motor neuron survival. LSM4 also associates with cancerous conditions as improper mRNA processing can lead to oncogene activation influencing processes like cell proliferation and apoptosis.
