Lyar also known as the cell growth-regulating nucleolar protein is a zinc finger protein with a molecular mass of approximately 36 kDa. This protein is expressed primarily in the nucleolus although it can also be found in other sub-nuclear structures. Lyar binds to nucleic acids and proteins playing roles in nucleolus-related processes. Its expression is seen in various tissues but it is most pronounced in rapidly dividing cells such as those in fetal tissues and certain cancers.
Biological function summary

The Lyar protein plays a role in regulating cell growth and proliferation. It participates in ribosome biogenesis by interacting with pre-ribosomal RNA possibly aiding in the stabilization and processing of rRNA precursors. Lyar is not usually considered as part of a protein complex but its interactions with several other nucleolar proteins imply its involvement in complex regulatory networks. It influences the transcription of ribosomal RNA genes impacting protein synthesis rates and cell cycle progression.

Pathways

Lyar is involved in the modulation of pathways linked to cell proliferation and survival. Notably it affects the PI3K/AKT signaling pathway by interacting with nucleophosmin (NPM1) impacting cell growth and survival mechanisms. This collaboration can alter the response of cells to external growth signals influencing cellular responses to metabolic changes and stress.

Aberrant Lyar levels have been implicated in oncogenesis. Its overexpression has been noted in various cancers including leukemia and neuroblastoma linking it to uncontrolled cell division and tumor progression. In cancer Lyar interacts with tumorigenesis-associated proteins such as c-Myc enhancing their transcriptional activity and contributing to cancerous transformation. Plays a role in the maintenance of the appropriate processing of 47S/45S pre-rRNA to 32S/30S pre-rRNAs and their subsequent processing to produce 18S and 28S rRNAs (PubMed : 24495227). Also acts at the level of transcription regulation. Along with PRMT5, binds the gamma-globin (HBG1/HBG2) promoter and represses its expression (PubMed : 25092918). In neuroblastoma cells, may also repress the expression of oxidative stress genes, including CHAC1, HMOX1, SLC7A11, ULBP1 and SNORD41 that encodes a small nucleolar RNA (PubMed : 28686580).
