Supplementary information

Lyl1 also known as Lim domain only protein 1 is a transcription factor with a mass of approximately 14 kDa. It plays a critical role in gene expression regulation. Lyl1 is expressed in hematopoietic tissues particularly in the bone marrow and lymphoid organs. It is known for its involvement in the development of certain lineages of blood cells. As a member of the basic helix-loop-helix (bHLH) family Lyl1 closely relates to other transcription factors that influence cellular differentiation.
Biological function summary

Lyl1 influences various cellular processes including the transcriptional regulation of hematopoietic stem cells. The protein does not typically function as part of a larger complex but interacts with core transcription machinery to modulate gene expression. Lyl1 affects lineage commitment and is important for the development of several types of blood cells especially impacting progenitor cells. Its presence and activity impact how blood cells multiply and differentiate into mature forms.

Pathways

The function of Lyl1 integrates into the hematopoietic and immune system pathways. It interacts with other proteins like Tal1 and HEB which are also bHLH transcription factors involved in blood cell development. Within these pathways Lyl1 contributes to the regulation and maintenance of stem cells ensuring balanced lineage differentiation and functional diversity within the hematopoietic system.

Aberrations in Lyl1 expression or function can lead to various forms of leukemia especially acute lymphoblastic leukemia (ALL). Lyl1 involvement in leukemia often connects with other oncogenes and regulatory proteins such as Notch1 which participate in the development and progression of the disease. Another disease associated with Lyl1 is lymphoblastic lymphoma where it contributes to uncontrolled proliferation and survival of affected cells. Understanding the specific role of Lyl1 in these disorders aids in developing targeted therapeutic strategies.
