The LYST protein also known as Lysosomal Trafficking Regulator or Beige is involved in intracellular transport processes. With an approximate molecular mass of 429 kDa it plays a role in the trafficking of lysosomal contents. Expressed in various tissues it particularly affects those with a high demand for lysosomal activity such as immune system cells. As part of its mechanical function LYST regulates the morphology of lysosomes and endosomes by facilitating fusion and movement of these vesicles.
Biological function summary

LYST directly influences the cellular degradation and recycling process. The protein does not act alone but functions as part of a larger cellular machinery that maintains lysosomal homeostasis. Its role is particularly important in immune cell function where it ensures proper lysosomal delivery and secretion especially in cytotoxic T cells and natural killer cells. These activities are important for granulocyte and lymphocyte regulation ensuring a proper response to infections and cellular stress.

Pathways

The protein LYST is integral in pathways related to vesicular transport such as the endocytic and autophagic pathways. It has a well-documented interaction with proteins such as Rab7 which is important for endosome-to-lysosome trafficking. Additionally LYST interacts with components of the cytoskeleton which further influences lysosome positioning within the cell therefore supporting its central role in cellular logistic pathways.

Mutations or functional disruptions of the LYST protein are linked to Chediak-Higashi Syndrome a rare genetic disorder characterized by immunodeficiency bleeding issues and neurological problems. This condition demonstrates a connection between LYST and the immune response as well as interactions with proteins like Perforin which are also involved in immune functions. Moreover LYST alterations might contribute to pigmentary anomalies in certain skin disorders due to its involvement in melanosome biogenesis.
