MACF1 also known as Microtubule Actin Cross-linking Factor 1 or ACF7 is a large protein with a mass of approximately 615 kDa. This protein serves as a cytoskeletal crosslinker playing an essential role in linking microtubules and actin filaments. MACF1 localizes to the cytoplasm and is expressed in various tissues throughout the body including muscle and brain. Its ability to interact with both microtubules and actin is central to maintaining cellular structure and dynamics.
Biological function summary

MACF1 facilitates cellular processes related to mechanical stability and intracellular transport. It integrates into diverse macromolecular assemblies including complexes that regulate cell polarization and migration. By influencing the architecture and dynamics of the cytoskeleton MACF1 contributes to the mechanical resilience and motility of cells. This protein also interacts with other cytoskeletal regulators supporting its role in dynamic cellular environments.

Pathways

There are several that MACF1 is actively involved with that intricately control cytoskeletal dynamics. It plays a major part in the Wnt signaling pathway interacting with proteins like GSK-3β to influence cell fate and development. MACF1 also crosslinks with the Hippo signaling pathway important for the regulation of organ size and cell proliferation. These pathways show how MACF1 intersects with key regulatory axes that coordinate cell movement and stability.

Mutations or dysfunctions in MACF1 often link to neurological disorders and muscular dystrophies. In Parkinson's disease disruptions in MACF1-associated signaling pathways might affect neuronal stability and survival. In muscular dystrophy defective association between MACF1 and dystrophin complicates muscle cell integrity. The impaired interactions between MACF1 and these associated proteins can profoundly impact tissue function and disease progression.

Isoform 2. F-actin-binding protein which plays a role in cross-linking actin to other cytoskeletal proteins and also binds to microtubules (PubMed : 16815997, PubMed : 18854161, PubMed : 21295697). Plays an important role in ERBB2-dependent stabilization of microtubules at the cell cortex (By similarity). Acts as a positive regulator of Wnt receptor signaling pathway and is involved in the translocation of AXIN1 and its associated complex (composed of APC, CTNNB1 and GSK3B) from the cytoplasm to the cell membrane (PubMed : 16815997). Has actin-regulated ATPase activity and is essential for controlling focal adhesions (FAs) assembly and dynamics (PubMed : 18854161). Interaction with CAMSAP3 at the minus ends of non-centrosomal microtubules tethers microtubules minus-ends to actin filaments, regulating focal adhesion size and cell migration (By similarity). May play role in delivery of transport vesicles containing GPI-linked proteins from the trans-Golgi network through its interaction with GOLGA4 (By similarity). Plays a key role in wound healing and epidermal cell migration (PubMed : 21295697). Required for efficient upward migration of bulge cells in response to wounding and this function is primarily rooted in its ability to coordinate microtubule dynamics and polarize hair follicle stem cells (PubMed : 21295697). As a regulator of actin and microtubule arrangement and stabilization, it plays an essential role in neurite outgrowth, branching and spine formation during brain development (PubMed : 26526844).
