Lanes 1 - 4 : Merged signal (red and green). Green - ab150371 observed at 24 kDa. Red - loading control, ab8245, observed at 37 kDa.

ab150371 was shown to specifically react with in wild-type HAP1 cells as signal was lost in MAD2L1 knockout cells. Wild-type and MAD2L1 knockout samples were subjected to SDS-PAGE. The membrane was blocked with 3% NF Milk. ab150371 and ab8245 (Mouse anti GAPDH loading control) were incubated overnight at 4°C at 1/10000 dilution and 1/20000 dilution respectively. Blots were developed with Goat anti-Rabbit IgG H&L (IRDye® 800CW) preabsorbed ab216773 and Goat anti-Mouse IgG H&L (IRDye® 680RD) preabsorbed ab216776 secondary antibodies at 1/20000 dilution for 1 hour at room temperature before imaging.

Mad2L1 also known as MAD2 or Kirk Mad2 is an important player in the mitotic checkpoint ensuring proper chromosomal segregation during cell division. It acts as a spindle assembly checkpoint protein weighing approximately 24.5 kDa. It is widely expressed in tissues with high proliferative activity including testis thymus and fetal liver. In these locations Mad2L1 binds to other mitotic checkpoint proteins to control the metaphase-anaphase transition.
Biological function summary

Target data

Component of the spindle-assembly checkpoint that prevents the onset of anaphase until all chromosomes are properly aligned at the metaphase plate (PubMed : 15024386, PubMed : 29162720). In the closed conformation (C-MAD2) forms a heterotetrameric complex with MAD1L1 at unattached kinetochores during prometaphase, the complex recruits open conformation molecules of MAD2L1 (O-MAD2) and then promotes the conversion of O-MAD2 to C-MAD2 (PubMed : 29162720). Required for the execution of the mitotic checkpoint which monitors the process of kinetochore-spindle attachment and inhibits the activity of the anaphase promoting complex by sequestering CDC20 until all chromosomes are aligned at the metaphase plate (PubMed : 10700282, PubMed : 11804586, PubMed : 15024386).
