MafB also known as V-maf musculoaponeurotic fibrosarcoma oncogene homolog B acts as a transcription factor involved in the regulation of gene expression. The protein has a molecular weight of approximately 37 kDa. MafB localizes predominantly to the nucleus where it exerts its function. Expression of MafB occurs in various tissues with significant levels in hematopoietic cells kidney and pancreas reflecting its role in cell differentiation and organ development.
Biological function summary

MafB plays an important role in the regulation of monocyte and macrophage differentiation. It serves as an important regulator that controls genes required for the immune response and terminal differentiation of these cells. MafB often operates as a heterodimer pairing with other transcription factors like Fos and Jun to form complexes that specifically bind to Maf recognition elements in DNA. This binding allows it to orchestrate various transcriptional programs necessary for immune system function and organogenesis.

Pathways

MafB integrates into the larger network of macrophage differentiation and the insulin signaling pathway. It interacts closely with proteins like PU.1 in the macrophage differentiation pathway where it helps specify the lineage commitment of hematopoietic progenitor cells. In the insulin signaling pathway MafB influences genes involved in pancreatic beta-cell function and glucose homeostasis showing its interconnected role in metabolic processes.

MafB connects to conditions such as diabetes and systemic lupus erythematosus (SLE). In diabetes MafB affects insulin secretion by regulating pancreatic beta-cell development and function and studies link its dysfunction to both Type 1 and Type 2 diabetes. In SLE aberrant expression of MafB possibly associates with autoimmunity where it might alter the normal immune response. Proteins like IL-6 involved in inflammation and immune response often interact with MafB highlighting the protein's significance in disease contexts.

Acts as a transcriptional activator or repressor (PubMed : 27181683). Plays a pivotal role in regulating lineage-specific hematopoiesis by repressing ETS1-mediated transcription of erythroid-specific genes in myeloid cells. Required for monocytic, macrophage, osteoclast, podocyte and islet beta cell differentiation. Involved in renal tubule survival and F4/80 maturation. Activates the insulin and glucagon promoters. Together with PAX6, transactivates weakly the glucagon gene promoter through the G1 element. SUMO modification controls its transcriptional activity and ability to specify macrophage fate. Binds element G1 on the glucagon promoter (By similarity). Involved either as an oncogene or as a tumor suppressor, depending on the cell context. Required for the transcriptional activation of HOXB3 in the rhombomere r5 in the hindbrain (By similarity).
