MAGEA9 also known as Melanoma-associated antigen family A 9 is a protein encoded by the MAGEA9 gene. This protein has a molecular mass of approximately 36 kDa. MAGEA9 is mainly expressed in testis and certain tumors making it a cancer/testis (CT) antigen. The specificity of this expression pattern makes MAGEA9 a valuable target in cancer research especially for immunotherapy approaches.
Biological function summary

MAGEA9 functions as an element facilitating cellular processes like proliferation and apoptosis. Although MAGEA9 itself is not part of any known complex its expression can influence the regulatory mechanisms within cells supporting cellular survival and growth under oncogenic stress. It has roles in modifying the transcriptional activity affecting genes involved in cell cycle regulation.

Pathways

The involvement of MAGEA9 extends to the p53 pathway and the MAPK signaling pathway both critical for cell cycle and survival regulation. In the p53 pathway MAGEA9 can interfere with normal p53 function which is vital for DNA damage response and apoptosis. MAGEA9 may interact indirectly with proteins such as p21 a well-known downstream effector of p53 impacting the pathway's ability to manage cell cycle arrest and maintain genomic stability.

MAGEA9 holds significant relevance in cancers particularly melanoma and lung cancer. The aberrant expression of MAGEA9 in these diseases highlights its potential utility in diagnostic and therapeutic applications. MAGEA9 expression has been linked with poor prognosis due to its potential to alter pathways related to cell death and proliferation. In some contexts MAGEA9 possibly interacts with other proteins like MAGEA4 which may collectively contribute to oncogenesis and tumor progression.
