Mammaglobin A also known as MGB1 is a breast-specific protein of approximately 93 amino acids with a molecular mass of about 15 kDa. It has a high expression in mammary tissue and limited expression in other tissues like the sweat glands. Researchers use immunohistochemistry (IHC) to detect Mammaglobin in tissue samples because of its tissue-specific expression making it a useful marker for breast epithelium.
Mammaglobin A functions as a glycoprotein. Its precise biological role remains unclear but scientists propose it plays a role in secretory processes due to its presence in epithelial cells of the mammary glands. This protein itself is not known to participate in a larger complex. The significance of Mammaglobin A as a marker rather than an active enzyme or receptor suggests its potential involvement in processes specific to breast tissue.
Mammaglobin A interacts within pathways involving sex steroid hormone signaling reflecting its expression in mammary tissue. This protein's expression is possibly regulated by hormonal pathways like the estrogen pathway which plays a significant role in breast tissue development and differentiation. Z- and S- Score: The Z-score represents the strength of a signal that a monoclonal antibody (MAb) (in combination with a fluorescently-tagged anti-IgG secondary antibody) produces when binding to a particular protein on the HuProtTM array. Z-scores are described in units of standard deviations (SD's) above the mean value of all signals generated on that array. If targets on HuProtTM are arranged in descending order of the Z-score, the S-score is the difference (also in units of SD's) between the Z-score. S-score therefore represents the relative target specificity of a MAb to its intended target.
A MAb is specific to its intended target if the MAb has an S-score of at least 2.5. For example, if a MAb binds to protein X with a Z-score of 43 and to protein Y with a Z-score of 14, then the S-score for the binding of that MAb to protein X is equal to 29.
