Anti-MAP3K8/COT antibody [RP23040075] is a Rabbit Recombinant Multiclonal antibody that is used in MAP3K8/COT ICC/IF, Western Blot. Suitable for Human, Mouse samples.

MAP3K8 is protein kinase which activated by phosphorylation during the S and G2/M phases of the cell cycle. MAP3K8 has been identified as an oncogene and can activate both MAP kinase and JNLK kinase pathways.

Immunofluorescence analysis of 4% paraformaldehyde on 70% confluent log phase U-87MG cells  labelling COT/MAP3K8 with ab308011 at 2 µg/mL, followed by Goat anti-Rabbit IgG (H+L) Secondary Antibody, Alexa Fluor® 488 conjugate at a dilution of 1/2000 (Panel a : green). Nuclei (Panel b : blue) were stained with DAPI. F-actin (Panel c : red) was stained with Alexa Fluor® 555 Rhodamine Phalloidin at 1 : 300 dilution. Panel d is a merged image showing cytoplasmic localization. Panel e is a no primary antibody control. The images were captured at 60X magnification.

Western blot analysis of MAP3K8/COT in whole cell extracts from Jurkat cells (lane 1), 3T3L1 cells (lane 2), and HepG2 cells (lane 3) using ab308011 at a dilution of 2 µg/mL. Samples were detected using chemiluminescence (ECL). Results show a band at ~52kDa.

Target data

Required for lipopolysaccharide (LPS)-induced, TLR4-mediated activation of the MAPK/ERK pathway in macrophages, thus being critical for production of the pro-inflammatory cytokine TNF-alpha (TNF) during immune responses. Involved in the regulation of T-helper cell differentiation and IFNG expression in T-cells. Involved in mediating host resistance to bacterial infection through negative regulation of type I interferon (IFN) production. In vitro, activates MAPK/ERK pathway in response to IL1 in an IRAK1-independent manner, leading to up-regulation of IL8 and CCL4. Transduces CD40 and TNFRSF1A signals that activate ERK in B-cells and macrophages, and thus may play a role in the regulation of immunoglobulin production. May also play a role in the transduction of TNF signals that activate JNK and NF-kappa-B in some cell types. In adipocytes, activates MAPK/ERK pathway in an IKBKB-dependent manner in response to IL1B and TNF, but not insulin, leading to induction of lipolysis. Plays a role in the cell cycle. Isoform 1 shows some transforming activity, although it is much weaker than that of the activated oncogenic variant.
