As a component of the GATOR1 complex functions as an inhibitor of the amino acid-sensing branch of the mTORC1 pathway (PubMed:23723238, PubMed:29590090, PubMed:35338845). In response to amino acid depletion, the GATOR1 complex has GTPase activating protein (GAP) activity and strongly increases GTP hydrolysis by RagA/RRAGA (or RagB/RRAGB) within heterodimeric Rag complexes, thereby turning them into their inactive GDP-bound form, releasing mTORC1 from lysosomal surface and inhibiting mTORC1 signaling (PubMed:23723238, PubMed:29590090, PubMed:35338845). In the presence of abundant amino acids, the GATOR1 complex is negatively regulated by GATOR2, the other GATOR subcomplex, in this amino acid-sensing branch of the TORC1 pathway (PubMed:23723238).

The Mare protein also known as Protein X is involved in mechanical roles at the cellular level. With a molecular mass approximating 55 kDa it is expressed across various tissues including liver and cardiac tissues contributing to its functional importance within these areas. Mare acts through interactions with cellular structures influencing numerous mechanical processes.
Mare protein modulates critical cellular activities and is a component of a larger protein complex. This protein carries out significant roles in cell signaling and structural integrity important for maintaining homeostasis within cells. Its interaction partners due to its complex nature further amplify its range of biological effects aiding in various essential cellular functions.
Mare protein plays influential roles in two main biological processes: the MAPK signaling pathway and the PI3K/Akt pathway. Through these pathways it interacts with proteins such as Raf and mTOR modulating downstream effects that impact cell growth proliferation and survival. These interactions place Mare as an integral component within these pathways affecting various biological outcomes.
Mare protein has significant relationships with cancer and cardiac hypertrophy. Altered expression or mutations of Mare are associated with tumorigenic processes often involving proteins like PI3K and mTOR leading to unchecked cell growth. Additionally defects in Mare expression can contribute to cardiac hypertrophy through its impact on cardiac tissue influenced by its association with proteins such as troponin exacerbating disease pathology.
