ab197590 staining MBNL3 in JAR (human choriocarcinoma) cells by ICC/IF (Immunocytochemistry/immunofluorescence). Cells were fixed with 4% Paraformaldehyde and permeabilized with 0.1% Triton X-100. Samples were incubated with primary antibody at a dilution of 1/500. A goat anti rabbit IgG (Alexa Fluor® 488) (ab150077) was used as the secondary antibody at a dilution of 1/1000. DAPI was used as a nuclear counterstain.

Muscleblind-like protein 3 (MBNL3) mechanically plays a role in RNA splicing and localization. It lacks a popular alternate name in scientific literature. MBNL3 has a mass of 36 kDa and is expressed widely in human tissues with higher expression in skeletal muscle heart and brain. The protein binds RNA via its zinc-finger domains enabling it to influence the processing of pre-mRNA transcripts.
Biological function summary

In the regulation of gene expression networks MBNL3 modulates alternative splicing and RNA stability. Though not part of a larger multiprotein complex it interacts directly with precursor mRNA specifically affecting splicing events. This regulation is critical in maintaining normal tissue function and development particularly in muscle and neural tissues.

Pathways

MBNL3 impacts RNA processing and stress response pathways. MBNL3 interacts with its homologs MBNL1 and MBNL2 which are part of the same splicing regulatory network. Through these interactions it affects pathways involved in cell differentiation and response to environmental stress contributing to the regulation of gene expression under varying physiological conditions.

Defects in MBNL3 function are associated with myotonic dystrophy and certain cancers. In myotonic dystrophy imbalanced splicing caused by malfunction of MBNL proteins including MBNL3 leads to muscular and neurological symptoms. In cancers changes in the expression or activity of MBNL3 might influence tumor progression through its role in splicing regulation. Understanding the connection between MBNL3 and diseases can inform therapeutic target research and potential treatment strategies.

Mediates pre-mRNA alternative splicing regulation. Acts either as activator or repressor of splicing on specific pre-mRNA targets. Inhibits cardiac troponin-T (TNNT2) pre-mRNA exon inclusion but induces insulin receptor (IR) pre-mRNA exon inclusion in muscle. Antagonizes the alternative splicing activity pattern of CELF proteins. May play a role in myotonic dystrophy pathophysiology (DM). Could inhibit terminal muscle differentiation, acting at approximately the time of myogenin induction.
