MCJ also known as DNAJC15 is a mitochondrial protein with an approximate mass of 17 kDa. It serves various functions via its presence in different tissues including the liver and muscle. MCJ belongs to the J-protein family and it regulates protein folding and mitochondrial function. Structurally MCJ acts as a co-chaperone and influences mitochondrial respiration by inhibiting the mitochondrial respiratory chain complex I.
Biological function summary

The role of MCJ extends beyond its mechanical actions. The protein participates in an intricate process that affects the cell's metabolism and energy production. MCJ often functions as part of a complex involving several mitochondrial proteins. It regulates mitochondrial energy production by inhibiting the activity of respiratory chain complexes leading to a direct impact on ATP synthesis and cellular metabolism.

Pathways

MCJ influences mitochondrial bioenergetics and oxidative stress pathways. It is intertwined with the mitochondrial respiratory chain pathway where it modulates the activity of complex I thereby affecting energy production and cell survival. MCJ interacts with other proteins such as NDUFA9 a component of complex I and plays a role in maintaining mitochondrial efficiency and reducing oxidative damage under stress conditions.

MCJ is associated with cancer and metabolic syndrome. Decreased expression of MCJ has been linked with various cancers due to its impact on cell survival and proliferation. In metabolic disorders alterations in MCJ expression affect energy metabolism and insulin sensitivity. Related proteins like the anti-apoptotic Bcl-2 family can further connect MCJ to these conditions illustrating the protein’s influence on disease-related mitochondrial dysfunction.
