Supplementary information

MCM10 also known as Mini-Chromosome Maintenance 10 is a component of the DNA replication machinery. This protein has an approximate molecular mass of 98 kDa. It functions mainly in the nucleus and is expressed across various cell types. MCM10 interacts closely with other replication factors to ensure DNA replication progression. Its mechanical role involves stabilizing the replication fork and binding to the replication origins.
Biological function summary

MCM10 plays an important role in the initiation and elongation phases of DNA replication. It acts as a scaffold for the assembly of other replication machinery components and facilitates the recruitment of DNA polymerase alpha. MCM10 is part of the larger pre-replication complex which includes other essential proteins such as MCM2-7 Cdc45 and GINS complex. These interactions ensure accurate DNA replication and cell cycle progression.

Pathways

MCM10 integrates into the DNA replication and repair pathways. Its function is critical in maintaining DNA integrity during the S phase of the cell cycle. In the DNA replication pathway MCM10's interaction with proteins like MCM2-7 and Cdc45 ensures the formation of replication forks. In the DNA damage response pathway it also plays a role by stabilizing the replication fork under stress conditions working alongside proteins like ATM and ATR to prevent replication stress.

MCM10 has links to cancer and genomic instability disorders. Abnormal expression levels of MCM10 can contribute to increased replication stress and genomic instability factors often seen in cancerous cells. MCM10 interacts with proteins like Cdt1 and Cdc6 whose dysregulation can lead to over-replication and cancer progression. Understanding MCM10’s role in these processes can provide insights into potential therapeutic interventions.

Target data

Acts as a replication initiation factor that brings together the MCM2-7 helicase and the DNA polymerase alpha/primase complex in order to initiate DNA replication. Additionally, plays a role in preventing DNA damage during replication. Key effector of the RBBP6 and ZBTB38-mediated regulation of DNA-replication and common fragile sites stability; acts as a direct target of transcriptional repression by ZBTB38 (PubMed : 24726359).
