The protein Microcephalin 1 (MCPH1) also known as BRIT1 plays an important role in cellular processes. It has a molecular mass of approximately 104 kDa. MCPH1 is widely expressed in various tissues with notable expression in the brain and gonads. Its primary mechanical function involves DNA damage response where it helps maintain genome integrity by recruiting repair proteins to sites of damage.
Biological function summary

MCPH1 acts as an important regulator in cell cycle checkpoints particularly the G2/M checkpoint which ensures that cells do not undergo mitosis with damaged DNA. This protein can form complexes with several others to effectively control the replication and repair processes. MCPH1 positively influences chromatin remodeling aiding in both the condensation and decondensation of chromatin structures during different phases of the cell cycle.

Pathways

MCPH1 integrates into both the ATM/ATR and CHK1/CHK2 pathways which are important in DNA damage signaling. These pathways involve the ATM protein which is related to MCPH1 through DNA damage response. In these pathways MCPH1’s regulatory actions are critical for proper AT response and checkpoint activation demonstrating its significance in maintaining genomic stability and cellular function.

MCPH1 mutations have been linked to microcephaly a developmental disorder characterized by reduced brain size. Additionally research associates MCPH1 dysfunction with cancer progression due to its role in maintaining chromosomal stability. The protein p53 connects to MCPH1 through both microcephaly and cancer pathways as it acts in concert with MCPH1 to promote accurate DNA repair and cell cycle regulation.
