Interpolated concentrations of native MeCP2 in human control wild type HAP1 (human near-haploid cell) and MECP2 (MeCP2) knockout HAP1 cell based on 250 ug/mL extract loads. The concentrations of MeCP2 were measured in duplicate and interpolated from the MeCP2 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean MeCP2 concentration was determined to be 1129.4 pg/mL in wild type HAP1 extract and undetectable in MECP2 (MeCP2) knockout HAP1 extract.

Target data

Chromosomal protein that binds to methylated DNA. It can bind specifically to a single methyl-CpG pair. It is not influenced by sequences flanking the methyl-CpGs. Mediates transcriptional repression through interaction with histone deacetylase and the corepressor SIN3A. Binds both 5-methylcytosine (5mC) and 5-hydroxymethylcytosine (5hmC)-containing DNA, with a preference for 5-methylcytosine (5mC).
