MED29 also known as Med2 is a component of the Mediator complex a multi-subunit assembly essential for the transcription regulation of RNA polymerase II. MED29 has an approximate mass of 29 kDa. It is expressed in various human tissues with high levels noted in brain heart and kidney tissues. The protein functions by acting as a coactivator modulating signals between transcriptional activators and the RNA polymerase II machinery. This allows MED29 to play a significant role in the control of gene expression.
Biological function summary

MED29 contributes to cellular processes through its role in the Mediator complex. This complex serves as a bridge facilitating communication between gene-specific transcription factors and the polymerase core machinery influencing transcription initiation. MED29 impacts the regulation of genes linked to cell growth development and stress responses. Given its participation in these important processes MED29 ensures that transcription responds accurately to changes in the cellular environment.

Pathways

MED29 plays a significant role in transcription regulatory pathways particularly the RNA polymerase II transcription pathway. MED29 interacts closely with other Mediator complex proteins such as MED1 and MED14 to drive precise gene transcription. These interactions facilitate a well-coordinated transcriptional response required for normal cellular functioning and response to stimuli.

MED29 has implications in cancer and cardiovascular disorders. Aberrant expression of MED29 links to its role in tumorigenesis supporting abnormal cell proliferation and survival. Similarly variations in MED29 expression impact heart function contributing to the development of cardiovascular diseases. MED29's association with other Mediator components such as MED12 further highlights its involvement in these conditions signifying its potential as a therapeutic target.

Target data

Component of the Mediator complex, a coactivator involved in the regulated transcription of nearly all RNA polymerase II-dependent genes. Mediator functions as a bridge to convey information from gene-specific regulatory proteins to the basal RNA polymerase II transcription machinery. Mediator is recruited to promoters by direct interactions with regulatory proteins and serves as a scaffold for the assembly of a functional preinitiation complex with RNA polymerase II and the general transcription factors.
