Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-MEIS1 antibody (AB19867)

IHC image of MEIS1 staining in Rat normal brain formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab19867, 0.5μg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

MEIS1 known as myeloid ecotropic viral integration site 1 is a homeobox protein that functions as a transcription factor. MEIS1 has a molecular mass of approximately 48 kDa. This protein is expressed in various tissues with high levels noted in the hematopoietic system particularly in progenitor cells. Its role in niave to the human developmental system is well detailed showing influence in developmental processes through gene expression regulation.
Biological function summary

As part of a protein complex MEIS1 interacts with other transcription factors including PBX1 and HOXA9 to modulate target gene expression. MEIS1 plays a critical role in hematopoiesis influencing the development and maintenance of hematopoietic stem and progenitor cells. Also it contributes to limb development and the formation of the heart during embryogenesis. MEIS1’s role in cellular differentiation and organogenesis highlights its significance in developmental biology.

Pathways

MEIS1 acts within the RA (Retinoic Acid) signaling pathway and is also linked to the Wnt signaling pathway. Through these pathways MEIS1 collaborates with other proteins like PBX1 which assist in the regulation of gene expression critical for proper cell fate determination. In the RA pathway MEIS1 modulates the transcription of genes necessary for cell differentiation and proliferation.
