Immunofluorescent analysis of 4% Paraformaldehyde-fixed, 0.1% Triton X-100 permeabilized IMR-32 (human neuroblastoma neuroblast) cells labellng MEIS1 with ab307544 at 1/100 dilution, followed by ab150081 Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) preadsorbed antibody at 1/1000 dilution (Green). Confocal image showing nuclear staining in IMR-32 cell line. Negative control : Daudi (PMID : 11094311). Image was taken with a confocal microscope (Leica-Microsystems, TCS SP8). is observed. ab195889 Anti-alpha Tubulin mouse monoclonal antibody - Microtubule Marker (Alexa Fluor® 594) was used to counterstain tubulin at 1/200 dilution (2.5 µg/ml) (Red). This data was developed using ab307544, the same antibody clone in a different buffer formulation. MEIS1 was immunoprecipitated from 0.35 mg IMR-32 (human neuroblastoma neuroblast) whole cell lysate 10 µg with ab307544 at 1/30 dilution (2µg in 0.35mg lysates). Western blot was performed on the immunoprecipitate using ab307544 at 1/1000 dilution. VeriBlot for IP secondary antibody(HRP)(ab131366) was used at 1/5000 dilution. Lane 1 : IMR-32 whole cell lysate 10 µg Lane 2 : ab307544 IP in IMR-32 whole cell lysate Lane 3 : Rabbit monoclonal IgG (ab172730) instead of ab307544 in IMR-32 whole cell lysate Blocking and dilution buffer and concentration : 5% NFDM/TBST. MEIS1 known as myeloid ecotropic viral integration site 1 is a homeobox protein that functions as a transcription factor. MEIS1 has a molecular mass of approximately 48 kDa. This protein is expressed in various tissues with high levels noted in the hematopoietic system particularly in progenitor cells. Its role in niave to the human developmental system is well detailed showing influence in developmental processes through gene expression regulation.
Biological function summary

As part of a protein complex MEIS1 interacts with other transcription factors including PBX1 and HOXA9 to modulate target gene expression. MEIS1 plays a critical role in hematopoiesis influencing the development and maintenance of hematopoietic stem and progenitor cells. Also it contributes to limb development and the formation of the heart during embryogenesis. MEIS1’s role in cellular differentiation and organogenesis highlights its significance in developmental biology.

Pathways

MEIS1 acts within the RA (Retinoic Acid) signaling pathway and is also linked to the Wnt signaling pathway. Through these pathways MEIS1 collaborates with other proteins like PBX1 which assist in the regulation of gene expression critical for proper cell fate determination. In the RA pathway MEIS1 modulates the transcription of genes necessary for cell differentiation and proliferation. These pathways demonstrate the broad regulatory influence of MEIS1 on cellular processes.

MEIS1 is linked with an increased risk for restless legs syndrome (RLS) and certain types of leukemia. The interaction of MEIS1 with HOXA9 is notably relevant in the pathogenesis of leukemia as this partnership can cause dysregulation in gene expression leading to oncogenesis. Additionally genetic variations near MEIS1 have been correlated with susceptibility to RLS highlighting its role in neurological conditions. This underlines the clinical importance of MEIS1 in both hematological and neurological disorders.
